Cascade catalytic platform modified intraocular lens for high-efficient posterior capsule opacification prevention

Abstract Posterior capsule opacification (PCO) is the commonest postoperative complication in cataract surgery, which is caused primarily by the proliferation, migration and differentiation of residual lens epithelial cells after intraocular lens (IOLs) implantation. Although recently emerging methods of IOLs surface modification have achieved certain effects for PCO prevention, there are still some problems, such as low long-term efficacy and poor intraocular biocompatibility, limiting its practical application significantly. This study reports on the introduction of a newfangled IOLs modified with double-enzyme triggered catalytic cascade reaction platform into ophthalmology for the efficient PCO suppression without using any toxic drugs. Natural glucose oxidase (GOD) and horseradish peroxidase (HRP) are simultaneously co-immobilized onto mesoporous channels of mesoporous silica nanoparticles (MSNs) to construct the cascaded catalytic platform (enzymes@MSNs), which was subsequently modified on polyethylenimine pre-coating IOLs to prepare enzymes@MSNs-IOLs. After modification, it still keeps excellent optical property and imaging quality, forecasting good application prospect. Comprehensive in vitro and in vivo assessments have demonstrated its high catalytic activities and significant PCO suppression. Therefore, this work not only introduces the concept of toxic drug free treatment into ophthalmology, but also achieves both desired PCO prevention efficacy and high intraocular biocompatibility.
