반응표면 분석법을 이용한 병아리콩 항산화활성 추출의 최적화 조건

This study was done to determine the optimum conditions from various solvent type(ethanol, methanol), solvent concentration(60, 70, 80%, v/v), extraction time(30, 80, 130 min.) and extraction temperature(10, 15, 20℃) in order to increase the antioxidant activity of the chickpea. DPPH radical scavenging activity was measured by using response surface methodology(RSM) to establish the optimal conditions. In the treatment of ethanol extractions, 80% of concentration for 130 min. at 20℃ showed maximum level of DPPH radical scavenging activity of 76.13%(R²=0.9435). However DPPH radical scavenging activity of methanol extraction was 73.28%(R²=0.9427) from 70% concentration at 20℃ for 130 min. The results indicated that ethanol extracts showed a higher antioxidant activity than that from methanol extracts. The ethanol extraction conditions of response surface analysis(RSA) were affected more by extraction time or concentration than solvent temperature(p<0.05). The methanol extraction conditions of RSA showed same resulted with the condition of ethanol extracts(p<0.001). Based on the RSM, the predicted best conditions of ethanol extraction were 72.88% of solvent concentration at 15.72℃ for 124.72 min. And also predicted best condition of methanol extraction were 70.16% of solvent concentration at 18.03℃ for 117.12 min.
