Expression and implication of ALS2CL gene in clear-cell renal cell carcinoma

Objective To study the differential expression of ALS2CL in clear cell renal cell carcinoma (ccRCC) and normal kidney tissue,and its implication.Methods Real-time polymerase chain reaction (PCR),immunohistochemistry and Western blotting were applied to examine the expression of ALS2CL in 26 cases of ccRCC tissues and adjacent normal kidney tissues as well as two kinds of ccRCCderived cell lines (786/O,SN12-PM6).Results As compared with normal control,the two ccRCC-derived cell lines (786/O,SN12-PM6) and 65.38% (17/26) of ccRCC tissues showed an obvious downregulated expression of ALS2CL mRNA.IPH revealed that 73.08% (19/26) of ccRCC specimens were negative or weakly positive for ALS2CL protein.Western blotting also showed a highly down-regulated rate of 53.85% (14/26) in ccRCC samples,and the low expression of ALS2CL was also found in 786/O and SN12-PM6 cells.All of them showed significant difference between ccRCC and corresponding normal control,P < 0.05.Conclusion The absence or downregulated expression of ALS2CL may be closely related to the occurrence of ccRCC.
