Growth and differentiation of HaCaT keratinocytes.

Abstract HaCaT cells are a spontaneously immortalized, human keratinocyte line that has been widely used for studies of skin biology and differentiation. Under typical culture conditions HaCaT cells have a partially to fully differentiated phenotype due to the high calcium content of both standard media and fetal bovine serum. This chapter describes low-calcium culture conditions for reverting HaCaT cells to the fully basal state followed by subsequent controlled differentiation using calcium induction.
