Human dermal fibroblast migration induced by fibronectin in autocrine and paracrine manners: Pilot study of remodeling in atopic dermatitis

-catenin were assessed using semi-quantitative RT-PCR,Western lot, or immunohistochemical methods. We found that of the Wnt amily members, Wnt5amRNA and protein levels were elevated in F as compared toNF. A higher expression of -catenin proteinwas lso found in KF. No detectable levels of FZD4 and ROR2 proteins ere observed in both NF and KF. Functional analysis showed that reatment of NF and KF with recombinant Wnt5a peptide resulted n an increase in protein levels of total -catenin and phosphoylated -catenin at Ser33/37/Thr41 but no significant change in hosphorylated -catenin at Thr41/Ser41 positions. In addition, he expression of total GSK3protein was not affected but its hosphorylated/inactivated form was increased in NF and KF. Our esults demonstrated a higher expression of Wnt5a and -catenin ut no detectable expressions of FZD4 and ROR2 receptors as well s inactivation of GSK3in KF as compared to NF. Taken together, hese findings highlight a potential role for aWnt/ -catenin canoncal signaling pathway triggered by Wnt5a in keloid pathogenesis. nt5a/ -catenin signaling pathwaymay provide a newmolecular arget for developing therapeutic strategies for keloid.
