The invention provides a fermentation medium of aureomycin, belonging to the technical field of fermentation culture. The fermentation medium comprises the following components by weight: 25-35g/L of glucose, 50-70g/L of vegetable oil, 30-50g/L of soybean cake meal, 5-7g/L of calcium carbonate, 20-30g/L of corn steep liquor, 4-6g/L of ammonium sulfate, 3-5g/L of sodium chloride, 0.2-0.3g/L of monopotassium phosphate and 0.2-0.3g/L of magnesium sulfate. The fermentation medium has the beneficial effects that the traditional concept in the fermentation field is broken through; plenty of oil is used in the fermentation medium, so that not only is the carbon source availability of strains increased but also the problem of foam generation in the fermentation process is solved and the fermentation titer is also substantially increased, specifically, the shake flask titer after aureomycin is fermented for 120 hours is more than 29000U/mL and the highest shake flask titer can be 31502U/mL.
