The time rule of pharmacological and toxicity effects of Cisplatin

Objective The research of time rule and relationship of pharmacological and toxicity effects of Cisplatin.Methods S180 tumor were transplanted to mice,the weight of tumor was weighed,blood BUN was determined by urinase UV velocity method,and LD50 of cisplatin in mice was evaluated in circadian time.Results Cisplatin used at 2:00,mice weight increase slightly,strong action,BUN was(28.1±4.5)mmol·L-1,strong toxicity,LD50 was(9.5±2.0)mg·kg-1;used at 18:00,mice weight increase a bit,weak action,BUN was(18.1±3.5)mmol·L-1,low toxicity,LD50 was(16.0±2.3)mg·kg-1.Conclusion The tumor inhibit effect,drug adverse reaction and toxicity of cisplatin may change synchronize with the circadian time.
