Determination of Eugenic Acid Content in Cornus officinalis Sieb. et Zucc. By HPCE

[Objective] The research was to develop an HPCE method for the determination of Eugenic acid Content in Cornus officinalis Sieb. et Zucc. [Method] Capillary column, uncoated capillary(50 cm×50 μm, effective length 40 cm); 1×TBE buffer solution; separated voltage, 20 kV; detection wavelength, 280 nm; temperature, 15 ℃; sampling conditions, pressure differential injection, 10 s; height: 8 cm. Before analyzing, capillary column was rinsed with 5 min with 0.1 mol/L sodium hydroxide solution, re-distilled water and buffer solution. Before sampling, 0.1 mol/L sodium hydroxide, distilled water and running buffer rinsed with 3 min. [Result] Eugenol had a good linear relationship with the peak area of integral linearity at the range of 0.08-1.00 mg/ml, the regression equation Y=19 760.03X-19.02(R2=0.990 3), the average recovery was 97.5%, RSD was 1.46%(n=6). [Conclusion] The method has many advantages, such as simple operation, accuracy and good repeatability, which is contributed to quality control of C. officinalis.
