Induction of callus from hypocotyl of Brassica napus L. and plant regeneration

High efficiency system of callus induction and plant regeneration in Brassica napus L.was studied.It would lay a foundation of further study on genetic transformation and somatic hybridization.Hypocotyl fragments were used as explants.The result showed that MS+2,4-D 2.0 mg/L was the best medium to induce embryonic callus,and the maximum inductivity was up to 90 percent.The callus can differentiate into plantlets when planted on MS+IAA 0.5 mg/L+6-BA 0.5 mg/L,and finally the plantlets can root on MS without additional growth regulators.Plant regeneration from callus via somatic embryogenesis was identified by histological observation.
