The present invention discloses a method using an enzyme - Chemical preparation of 1,3-Directed -2-oleic palmitic acid triglyceride, glycerol as a starting material first, under stirring, the temperature in the reaction with oleic acid dissolved, added free or immobilized lipase as catalyst, selective esterification, to give 1,3-position diglycerides; palmitic acid derivative and diglycerides palm then obtained or to obtain 1 , 3-oleic acid triglyceride palmitic -2- crude product; Finally, molecular distillation, to give 1,3-dioleate -2- palmitic triglyceride claim. Preparation of 1,3-DAG of the present invention by screening for specific lipases obtained 1,3-DAG content of more than 90%, more than 90% yield; finally obtained 1,3-oleic acid - glycerol diester content up to 95%, more than 95% yield.
