The influence of microenvironment on rooting of Lilium tsingtauense
2013
To obtain the optimum culture media for bulblets rooting of Lilium tsingtauense, the influences of the concentration of NAA,sucrose, activated charcoal and CCC on the rooting of Lilium tsingtauense in vitro microenvironment were studied. The results indicated that the sucrose could significantly influence the enlargement of blublets, the plantlets grow well when 60 g/L sucrose was added in the media;dark treatment or 3 g/L activated charcoal could build dark microenvironment for root growth, which was beneficial to promote the differentiation of root primordial so that the bulblets rooted faster, under which the roots emerged 2~3 d earlier than other treatments;2 g/L CCC could increase the dry weight, which enhanced the resistance of vitro shoots. It could obtain a number of vitro shoots quickly when the treatment was 1/2 MA+0.1 mg/L Naa+60 g/L sucrose+3 g/L activated charcoal or dark treatment;it could obtain strong vitro shoots when the treatment was 1/2 MS+0.1 mg/L NAA+60 g/L sucrose+2 g/L CCC as well as under illumination condition.
