A method for the production of regulatory T cells (Treg) reagents human donor, comprising: a) co-culturing CD19 + B lymphocytes of a donor hum five year with feeder cells human leukemia CD40L + irradiated under conditions effective to produce stimulated lymphocytes B ( SBC); b) co-culturing CD4 + T cells, CD25 +, CD127- / lo of a human recipient with said Sbc under conditions effective to expand selectively regulatory T (Treg) lymphocytes reactive human donor; c) restimulate said Treg crosslinking reagents donor by CD3 and CD28 Treg said reactive donor under conditions effective to produce Treg donor reagents that are CD4 + and Foxp3 + Helios +; wherein the donor is HLA mismatched relative to the human recipient.
