Effects of Gliocladium roseum Bain on adriamycin nephropathy in rats and its mechanism.

Objective To discuss the therapeutical effects and mechanisms of Gliocladium roseum Bain on the chronic kidney disease. Methods Twenty-four male Sprague-Dawley rats were randomly divided into normal control group(N group),model group(M group)and Gliocladium roseum Bain group(G group)with 8 in each. The nephropathy model were established in M and G group by two times injection of adriamycin (ADR)and unilateral nephrectomy,then the rats in G group were intragastriced with Gliocladium roseum Bain after the first injection of ADR,while the other two groups were treated with equivalent normal saline. The renal function and biochemical indicator of rats were detected at the weekend of 8th,the pathological changes of nephridial tissue were observed by HE and PAS stain methods,the relative quantity of molecular biological index of nephridial tissue were measured by fluorescent quantitation RT-PCR. Results Compared to the N group,the 24h urine protein ,blood fat ,SCr and BUN level in M group and G group were higher,the plasma-albumin level was lower ,and there were significant difference between the indexes in M group and G groups(P0.05,0.01). Pathological changes of nephridial tissue in N group were normal,while focal segmental glomerulosclerosis and significant inflammatory cell infiltration were found in M group,the changes in G group were slighter than that in M group,area of glomerulus ECM (M)/ area of glomerulus in M groupG groupN group,all P0.05.The expression of TGF-β1,TIMP-1,PAI-1 mRNA in G group were lower than those in M group(P0.05,0.01). Conclusion Gliocladium roseum Bain can significantly relieve the renal injury of ADR -induced nephropathy,the mechanisms maybe down-regulate the expression of TGF-β1,TIMP-1 and PAI-1.
