Cloning and Sequence Analysis of VP1 Gene of Avian Encephalomyelitis Virus Shaanxi Strain

In order to reveal the molecular characteristics of VP1gene of avian encephalomyelitis virus(AEV),the VP1gene of AEV Shaanxi strain was cloned and analysed.The VP1gene sequence of AEV Shaanxi strain was successfully amplified with a pair of specific primers by RT-PCR,then cloned into pMD18-T vector and sequenced.The sequence of VP1gene was compared with those of the NCBI published strains.The results showed that the full length sequence of VP1gene of SX strain consisted of 810 nucleotides,coding 270amino acid residues.The VP1gene sequence analysis showed that it shared 91.5%99.4% nucleic acid identities and 88.5%-98.9%amino acid identities;three transmembrane domains were located in the regions from 1to 7,from 19to 25,from 149to 155amino acids respectively.Evolution analysis indicated that SX strain was closely related to 1143and L2Zstrain.
