Transcript abundance of CATSPER 1 gene in spermatozoa of good and poor Quality Semen Producing Frieswal Crossbred Bull

The present research work deals with the expression profile of CATSPER-1 gene as it plays an important role in sperm function, specifically sperm motility, capacitating and acrosomal reaction. For this purpose, ejaculates were obtained from crossbred Frieswal bulls and assessed for normal semen quality parameters and thereafter categorized into Good and Poor quality semen groups based on initial progressive motility. Eosin nigrosin (EN) vital stain and hypo-osmotic swelling test (HOST) were performed for evaluation of the sperm plasma-lemma integrity. After sperm recovery by Percoll gradient centrifugation, total RNA extraction was done by TRI reagent method. Real-time qPCR was used to elucidate the differential expression of can-didate gene CATSPER 1. One-way ANOVA using SPSS program and Student t-test were performed to determine the significant differences of CATSPER 1 expression profile and comparison of means be-tween good and poor quality semen producers, re-spectively. The present study although revealed slightly lower expression of CATSPER1 in the sper-matozoa of normal group compared to the motility impaired group of crossbred Frieswal bulls, howev-er, no significant difference (P> 0.05) in transcript abundance was observed between the groups.
