Development of an optimized peptide map for recombinant activated human protein C by means of an experimental design strategy

Abstract An experimental design strategy was used to optimize the digestion conditions of a peptide mapping method for recombinant activated human protein C (aPC). Initially a Plackett-Burman screening experiment was used to identify important experimental parameters. Subsequently those parameters found to be important in the screening study were optimized using a full factorial experiment. The data obtained allowed selection of optimized digestion conditions which ensured complete tryptic digestion and minimized non-tryptic cleavages.
