Inhibition of proliferation of salivary adenoid cystic carcinoma by targeting gene transfer

目的 探讨人端粒酶逆转录酶(human telomerase reverse transcriptase,hTERT)启动子调控肿瘤坏死因子相关凋亡诱导配体(tumor necrosis factor-related apoptosis-inducing ligand,TRAIL)基因在涎腺腺样囊性癌细胞(salivary adenoid cystic carcinoma,SACC)-83中表达及对SACC-83增殖的影响.方法 脂质体介导pACTERT-TRAIL转染SACC-83和人胚肺成纤维细胞(human embryonic lungfibroblast,HEL),通过RT-PCR检测TRAIL表达,甲基噻唑基四唑法检测细胞存活率,流式细胞仪检测细胞凋亡率.结果 hTERT启动子调控TRAIL基因在SACC-83中明显表达,抑制其增殖(存活率62.80%),且诱导细胞凋亡比例增加(10.89%);而在HEL细胞中未能诱导TRAIL基因表达,对该细胞的增殖和凋亡无明显影响.结论 hTERT启动子可诱导TRAIL基因在SACC-83中特异性表达及诱导细胞凋亡。
