NOVEL SEQUENCING TECHNIQUES APPLIED TO THE “tRNA-LIKE” 3′-TERMINUS OF TURNIP YELLOW MOSAIC VIRUS RNA

ABSTRACT. A fragment representing the 3′-terminal “tRNA-like” region of Turnip Yellow Mosaic Virus RNA has been isolated, and partially sequenced. Sequence information was derived entirely by in vitro [ 32 P] end-labelling techniques. The new methodologies developed in the course of this work, particularly the use of nuclease P 1 in the generation of partial digests for direct sequence analysis by two-dimensional homochromatography, may prove of general utility in the sequence determination of 3′- and 5′-ends both of tRNA and of high molecular weight RNAs, such as viral and other mRNAs.
