The inhibiting effect of cisplatin on cells and telomerase activity of squamous cell carcinoma of tongue

Objective To study the inhibiting effect of cisplatin on human tongue Tca8113 cells and telomerase activity.Methods Tca8113 cells were treated in vitro with cisplatin and the cells untreated with cisplatin as controls.The inhibiting effect and the chang of telomerase activity induced by cisplatin in human tongue cells Tca8113 were detected by MTT assay in vitor,clonogenic assay,transmission electron microscope,flow cytometry analysis techinique and TRAP?PCR?ELISA.Results Activity of Tca8113 cells treated with cisplatin was significantly inhibited,that was density-dependent.Morphology of cell was changed but cell cycle was not changed.Telomerase activity of Tca8113 cells treated with cisplatin was significantly inhibited,and the effect was time-dependent.Conclusions Cisplatin can effectively inhibit the cell activity and inclination of transfer and recurrence of Tca8113 cells.The cispiatin can act on the mitochondrions of cells.It can refrain the telomerase activity and change the cell cycle little at the same time.
