MONITORING AND CONTROL OF HYBRIDOMA CELL LINE MN12 IN A CONTINUOUS PERFUSION CULTURE SYSTEM

Data are presented on cultivation of hybridoma cell line MN12 in a continuous culture system with cell retention. The ADI 1000 computerized measurement and control system has been used (1). Process parameters controled were pH, dissolved oxygen (DO), various gas flows, temperature and the amount of oxygen supplied to the headspace, which is a measure for the oxygen demand of the cells. During cultivation a change from aerobic to a more anaerobic metabolism was observed, which also affected the glycosylation pattern of the monoclonal antibody (Mab) as determined with lectins. ELISA's, isoelectric focussing (IEF) and FC-analysis were performed to characterize the amount and integrity of the produced Mab.
