Polyethylene glycol conjugated interferon-beta-1b with enhanced biological potency in vitro
2003
A process for increasing the in vitro biological potency of interferon-beta unglycosylated selectively comprendeacoplar in the presence of a surfactant, one or more soluble polymers synthetic water alaminoacido the amino terminus of said interferon-beta, wherein said amino acid amino estalocalizado remotely from (the) domain (s) receptor binding of the interferon-beta, and wherein (i) the water-soluble polymer bearing a single aldehyde group and the coupling comprises alkylation reductorade a Schiff base formed with the water-soluble polymer and reduction of the Schiff base with a soft agentereductor, or (ii) the coupling comprises coupling a hydrazide, hydrazine, semicarbazide or other soluble polymer aguaque amine containing a serine or threonine end N interferon-beta that has been escindidooxidativamente periodate to an aldehyde, yen which the water soluble polymer is a polialqui glycol selected from the group consisting of poly (ethylene glycol), monomethoxypoly (ethylene glycol) and monohidroxipoli (etinelglicol).
