Study on the Antitumor Activity of Protein Extracts of Peganalaum harmala Seeds in vitro

Objective:To evaluate antitumor efficacy of the protein extracts of Peganalaum harmala seeds in vitro.Method:The crude proteins of Peganum harmala L.seeds were extracted by saturated ammonium sulfate.Cell growth inhibitory effects of the crude protein extracts on tumor cells in vitro were examined with MTT assay.Cell apoptosis was detected by flow cytometry (FCM) and annexin V/PI fluorescence staining.Result:The 80% saturation of ammonium sulfate from Peganalaum harmala seeds protein had the cell inhibition activity against HeLa,Eca109,and BEL-7404 cells,IC50 were 72.11±3.88μg/mL,257.58±1.16μg/mL,and 174.07±1.32μg/mL,respectively.It showed significantly cell inhibition activity against HeLa cell (P0.01),and induced apoptosis in a dose-dependent manner.Conclusion:The Peganalaum harmala seeds protein could significantly inhibit HeLa growth and induce its apoptosis.
