Cloning and Sequence Analysis of GroEL Gene of Brucella ovis Strain 80/019 Isolated from Xinjiang

A pair of primers was designed and synthesized based on the nucleotide sequence of GroEL gene of brucella abortus.One target fragment about 1.6kb was amplified via PCR from the genomic DNA of Brucella ovis strain 80/019 isolated from Xinjiang with the designed specific primers.After purification,the target fragment was cloned into linearized plasmid vector PBS-T and sequenced.The ORF of GroEL gene was 1641bp,and the nucleotide sequence homology of GroEL gene of brucella ovis with that of brucella abrotus,brucella melitensis,brucella suis,was 99.88%,99.88%,(99.82%) respectively,and a high degree of amino acid sequence homology was revealed so the nucleotide sequence and amino acid of homology is the highest.
