The main function of the human sebaceous gland is sebum excretion. Increased sebum levels combined with follicular hyperkeratinization are prerequisites of acne vulgaris. Some transcription factors have been identified, such as peroxisome proliferator-activated receptors (PPARs), CCAAT/enhancer binding proteins (c/EBPs) and sterol regulatory element-binding protein 1 (SREBP-1), which are known to control lipid metabolism in adipocytes. In the current study, we investigated the anti-inhibitory effect of Graviola leaf extract (GLE) on lipid synthesis in human sebocytes. Treatment with GLE significantly suppressed lipid content of human sebocytes in a dose-dependent manner. The concentrations of cellular triglyceride and cholesterol were also reduced in human sebocyte after treatment with GLE when compared with the controls. In addition, GLE also reduced the expression levels of the transcription factors C/EBP-1, PPAR-γ and SREBP-1 significantly. Furthermore, GLE modulated the phosphorylation of ERK and Akt signal molecules. Taken together, these findings revealed that GLE showed strong anti-inhibitory effects on lipid synthesis in human sebocytes.
