Genetic diversity for the provenance of Picea koraiensis by RAPD markers

To reveal the genetic diversity of Picea koraiensis, 80 samples from eight provenances of this species in northeast part of China have been examined by using RAPD level. 17 loci have been gotten from 80 samples using 12 RAPD primers, in which 126 polymorphic loci can be seen. Proportion of the total polymorphic loci has been reached 73.68%. There are some changes in proportion of polymorphic loci among eight provenances, which is from 35.09% to 45.03%. Also, Shannon's Information Index and Nei's Gene Diversity Index are used to estimate genetic diversity for the provenances of Picea koraiensis, and there are almost the same varying trends for these two indexes among the provenances, they are Huanan (HN), Wuyiling (WYL), Chaihe (CH), Muling (ML), Tianqiaoling (TQL), Aihui (AH), Gaofeng (GF) and Dafeng (DF) from low to high in turn. The genetic differentiations for both of inter-provenance and intra-provenance are 42.26% and 57.74%, respectively. It also shows tht there is close relationship between different provenances because the mean genetic identity among the provenances is 0.8622, varying from 0.8046 to 0.09237. On the basis of cluster analysis of geneetic distance, eight provenances have been divided into three groups, in which the first group includes Tianqiaoling and Muling provenances; the second is Chaihe and Huanan provenances; and the last group contains Dafeng, Wuyiling, Aihui and Gaofeng provances. In addition, there exists a positive correlation between genetic distance and geographic distance among the provenances to a certain degree, and the correlation coefficient is 0.4394.
