Effects of Preliminary Treatment on Watermelon Anther Culture Callus Induction

The anthers of six watermelon genotypes were used as test materials to investigate the factors of the time of anther low temperature pretreatment and HgCl2 disinfection on watermelon anther culture callus induction. The results showed that the callus induction ratios were significantly different among tested watermelon genotypes,1005 had the highest callus induction rate;The pretreatment of anthers at 4 ℃ for 48 h and 72 h can induce higher callus rate,the callus induction ratios significantly higher than those of the control;When the time of the 75% alcohol sterilization was 30 s and 0.1% HgCl2 disinfection was 8 min,the pollution rate and browning rate of the watermelon anther were all lower,that can be used as main disinfection method of watermelon anther culture.
