Cellular mechanisms of lipophilic cationic drug distribution: a confocal laser microspectrofluorometric assessment

1995
ABSTRACT Ellipticine has been taken as a model for antitumoral compounds having weak base properties(7subcellular local pHs, as well as by the modification of the protonation equilibrium of the drug when bound to various biological macromolecules. INTRODUCTIONMost of the agents used in chemotherapy (such as antitumor drugs) as well as numerous fluorescent probes designed to monitor intracellular physiological parameters (pH or Ca++ indicators, potential sensitive probes, nucleic acid stains,.. .) are lipophilic compounds which usually bear a cationiccharge at a physiological pH. Their lipopilic character allows their passive diffusion accross biological membranes without involving carrier proteins. It also leads to hydrophobic interactions with some
