Expression of miRNA-149 in bladder cancer tissue and its biological significance

Objective To investigate the expression of miRNA-149 in bladder cancer and its biological significance. Methods The expression of miRNA-149 in paired tissues and bladder cancer cells was detected by qRT-PCR. The miRNA-149 over-expression plasmid transfected bladder cancer cells T24, T24T by PolyJet™DNA In Vitro Transfection Reagent, and the transfection rate was detected by fluorescence quantitative PCR. The migration and invasiveness of cells were detected by Transwell method. Results The expression of miRNA-149 in invasive T24 and T24T cells were significantly lower than that in bladder epithelial immortalized cells; in 14 matched patients, the expression of miRNA-149 in bladder cancer tissue was significantly lower than that in adjacent normal tissue, with statistically significant differences (P<0.05). In miRNA-149 stably over-expressed T24 and T24T cells, miRNA-149 could significantly inhibit the invasion of bladder cancer cells, but could not significantly inhibit the migration of bladder cancer cells, with statistically significant differences (P<0.05). Conclusions miRNA-149 is abnormally expressed in bladder cancer tissue and cells, and the up regulation of miRNA-149 may inhibit invasion of bladder cancer cells, which provides a new biological significance for the study on bladder cancer metastasis.
