An effective methodfortheisolation ofnucleoli from auxin-treated soybean(Glycine max,var.Wayne)hypocotyl was developed by polytronhiomogenization and sucrose gradient centrifugation. The nucleoli expressed onlythe a-amanitin-insensitive RNA synthetic activity. Thisactivitychromatographed asRNA polymerase IonDEAE-cellulose.Itappearsthattheplantnucleolus, liketheainimal nucleolus, isthesiteoflocalization forRNA polymerase I.
