Protection of Human Bone Microvascular Endothelial Cells from Glucocorticosteroid Induced Cell Injury by Icariin in vitro

Objective To establish the model of human bone microvascular endothelial cells (HBMECs) injury induced by glucocorticosteroid;To investigate the effect of icariin on human bone HBMECs injury induced by glucocorticosteroid. Methods HBMECs were isolated through immunomagnetic beads,cultured in vitro. The model of HBMEC injury induced by glucocorticosteroid was established. The cytotoxicity of HBMECs was measured by MTT assay. The injury of HBMECs induced by the glucocorticosteroid was detected by trypan blue stainning. Results Significant cytotoxicity of HBMECs was induced by glucocorticosteroi with dose dependent manner and time dependent manner;The necrosis of HBMECs after incubation with glucocorticosteroid was found by trypan blue staining. ICA could significantly inhibit the glucocorticosteroid induced necrosis of HBMECs. Conclusion ICA can protect the injury of HBMECs induced by glucocorticosteroid,our study provided some experimental pharmaco-foundation for intervention therapy of early osteonecrosis.
