Establishment of rats model with actue hepatic injury induced by injection of concanavalin A

Objective To establish the rats model with actue hepatic injury induced by injection of concanavalin A.Methods Sixty-nine BLAB/c mice were divided in five groups.Rats in group A were injected concanavalin A(15mg/kg).Rats in groups B,C and D were injected with anti-TNF-α and (or) anti-IFN-γ(20μg/mouse) at 30min before concanavalin A injection.The rats in group E were given normal saline as the controls.The blood samples were taken on the 4th,8th,and 12thh for detecting tumor necrosis factor(TNF)-α and interferon(IFN)-γ by ELISA and the liver was excised for HE staining and interleukin-18(IL-18) determination by Western blotting.Results At the end of experiment,the mice number of abnormal death in groups A,B,C,D and E was 6,2,3,1 and 0,respectively.A widespread hepatocellular necrosis was seen in group A on the 8th h,which was much lighter in groups B,C and D(P0.05). The expressions of TNF-α and IFN-γ were increased,which were decreased in groups B,C and D(P0.05). The expression of IL-18 was strongly positive. Conclution The rats model with actue hepatic injury induced by injection of concanavalin A has been established successfully.The formation of the model is related to TNF-α,IFN-γ and IL-18,which may be blocked by preinjection of anti-TNF-α and(or) anti-IFN-γ.
