The invention discloses a molecular detection method for related mutation for generating resistance on ACCase inhibitor herbicides by alopecurus japonicas based on a derived cleaved amplified polymorphic sequence (dCAPS) technique. The molecular detection method can be applied to quick and accurate detection of the drug-resistant related mutation of the alopecurus japonicas, and belongs to the technical field of organisms. The method comprises the following steps: respectively designing primers for different mutations according to the alopecurus japonicas and resistance-related ACCase gene; carrying out polymerase chain reaction (PCR) amplification and obtaining an amplification segment by taking genome deoxyribonucleic acid (DNA) of the alopecurus japonicas as a template; selecting corresponding restriction endonuclease to carry out digestion on the PCR amplification segment; and after digestion, judging whether the alopecurus japonicas has resistance-related mutation and judging genotype of the mutation according to the agarose gel electrophoresis. Compared with the traditional mutation detection method, the molecular detection method is strong in specificity, and high in accuracy, the method is faster and simpler, the detection cost is greatly reduced, and an excellent molecular detection method is provided for detection of the resistance-related mutation of the alopecurus japonicas.
