Construction of small interfering RNA expression vector targeting FoxM1 gene

Objective To construct a siRNA expressing vector targeting FoxM1,in order to investigate the effect of FoxM1 gene in lung cancer and other diseases.Methods A FoxM1 targeting short hairpin DNA fragment was designed and synthesized,and then was cloned into a linearized backbone plasmid pSilencer2.l-U6.The new plasmid was identified to be correct by cutting with restriction enzymes and gene sequencing.Results The FoxM1 targeting shDNA fragment was inserted into pSilencer 2.1-U6 backbone vector correctly.Conclusions The small interfering RNA expression vector targeting FoxM1 gene is constructed successfully.
