Method for high-speed chromatographic detection of hemoglobin variants and quantitation of hemoglobins and f a2.

1998
A method of analyzing several individual blood samples to determine the indicator characteristics in these a hemoglobin variant, and for quantifying the amounts of hemoglobin A2 and F in said samples, said method comprising the following steps: (a) analyzing a blood sample respective passage through an HPLC column, said HPLC column consisting essentially of a cation exchange column maintained above about 30C; (B) eluting the normal and abnormal hemoglobin species including Hb-of C with a current passing eluent through the HPLC column; (C) characterizing eluted species as a function of their respective absorbance values and retention times, said characterizing step the step of quantifying the amounts of A2 and F hemoglobins in the respective blood samples; and (d) repeating steps (ac) to analyze each of the multiple blood samples, wherein the total time between successive steps (ac) is up to about 5, 5 minutes.
