The protective effects of SB203580 on hematopoietic tissue exposed to irradiation

Purpose To investigate the protective effects of SB203580 on hematopoietic tissue of C57 BL/6 mice injured by γ-ray.Methods Male C57 BL/6 mice were randomly divided into control group(Ctr),irradiation group(IR) and SB203580 treatment group(SB).Mice were intraperitoneal injected SB(15 mg/kg)30 minutes ahead of TBI and then given every 2 days for5 times after the TBI.On day11,pe-ripheral blood count and nucleated femoral cell count were performed by blood analyzer.1 ×105 cells were seeded into Methocult GF M3534 methylcellulose medium and colony-forming units-granulocyte and mono-cyte(CFU-GM) were scored on day 6.DCFH as a fluorescent probe assessed the reactive oxygen species(ROS) generation in overnight incubated bone marrow mononuclear cells(BMMNC),which were irradiatedat 1 Gy.Results Transitory and significant changes occurred in the cell amount of BMMNC in the irradiated mice after administration of SB203580: white blood cells,platelets and BMMNC in SB were significantly in-creased by 111.8%,157.7% and 135.4%(P 0.05).The proliferation suppression of BMMNC in IRgroup was eased by SB203580 injection and CFU-GM in SB group score rises by 118.5%(P 0.05).More-over,ROS gets back to the normal level observed in SB group.Conclusion It was shown that changing withcell count and enhancing of proliferation manifested the protection of SB203580 from radiation induced hematopoietic tissue injury.The protection of SB203580 may attribute to its suppression on ROS generation.
