Morphological studies were used to evaluate the protective effect of afobazole on the experimental model of ischemic stroke caused by the middle cerebral artery occlusion (MCAO). It was found that afobazole injections at a dose of 5 mg/kg twice a day resulted in good safety of stryopellidar systum, the area of ischemic necrosis of the cortex was clearly limited and characterized by early reperfusion and good preservation of neurocytes of basal and hypothalamic nuclei, and decreased the severity of hypoxic damage to neurocytes in the cortex of fields Cg1 and Cg2. Afobazole exhibited neuroprotective activity both after preliminary treatment and after its injection within therapeutic window (after 6-h MCAO), which was manifested by the elimination of subarachnoidal edema, preservation of neurocytes in the perifocal zone, and revascularization of the ischemic zone. Thus, the obtained results demonstrated a pronounced neuroprotective activity of afobazole in ischemized brain.
