Benzeneborinic acid derivative, preparation thereof and use thereof as synthetic intermediate

1993
PURPOSE: To obtain a new benzeneborinic acid derivative which may be coupled with a halogenated aromatic compound or a halogenated heterocyclic ring containing a plurality of substituents. CONSTITUTION: The new compound is a compound of formula I (wherein R 1 -R 3 are each 1-2C alkyl or aryl; and -CR 1 R 2 R 3 is located at 1-position or 2-position of the tetrazole ring), e.g. bis[2-[2-(1,1-dimethylethyl)-2H-tetrazol-5- yl]phenyl]borinic acid. The compound is obtained by allowing 2-(1,1- dimethylethyl)-5-phenyltetrazole to react with an alkyl lithium in an aprotic solvent at -55 to 66°C to obtain an organic lithium compound, which is then allowed to react with a trialkyl borate in an aprotic solvent to obtain an alkyl benzeneborinic acid, and then hydrolyzing this. The compound of formula I is used as a synthetic intermediate for a compound of formula II (wherein Ar is aryl, naphthyl, pyridyl, pyrimidinyl, thienyl, imidazolylquinolyl or imidazopyridyl).
