Resonance Rayleigh Scattering Spectrometric Determination of Hemoglobin by its Reaction with CdTe Quantum Dots

In HAc-NaAc buffer(pH 6.0),hemoglobin reacted with CdTe quantum dots to form ionassociation complexes,which greatly enhanced the resonance Rayleigh scattering(RRS) intensity of the system at the wavelength of 460 nm.The optimum reaction conditions including pH,the concentration of CdTe quantum dots and the intensity of ionic of the system were investigated.Under optimum condition,the RRS intensity(ΔI) at 460 nm was linear to the concentration of hemoglobin in the range of 6.6~189.2 mg/L.The regression equation was △IRRS = 139.9 + 1.123 C,with a correlation coefficient of 0.992 8.The detection limit was 0.5 mg/L.Based on this,a novel method for the determination of trace amounts of hemoglobin by using the RRS method was developed and used for the analysis of human serum samples.The recoveries were in the range of 97.0%~103.4%,and the RSDs were 3.2%~6.1%.The proposed method was simple,sensitive and rapid.
