Fast determination of steroid hormones in fish blood with micellar electrokinetic capillary chromatography

A novel micellar electrokinetic capillary chromatography(MEKC) method was developed for separation and detection of cortisol,17 β-estradiol and testosterone in fish blood.The factors affecting separation efficiency such as concentration of buffer solution and surfactant,additives,pH,separation voltage and temperature were investigated.The optimal conditions were concluded as follows: 12 mmol/L borax+60 mmol/L sodium dodecyl sulfate(SDS)+6 mmol/L β-cyclodextrin(β-CD) + 8%(v/v) acetonitrile,pH 9.5,detection wavelength of 225 nm,separation voltage and temperature of 25 kV and 25 ℃,respectively.Under the optimal conditions,the analytes could be base-line separated within 4 min using reverse separation mode with capillary length of 10 cm.Good linear relationship were observed in the range of 1.0~100.0 μg/mL with correlation coefficient larger than 0.993 2.The proposed method had been successfully applied to the determination of steroid hormones in fish blood with recoveries in range of 84.60%~113.30%.
