FLOATING PULSATILE DRUG DELIVERY SYSTEM: A REVIEW

Recent studies reveal that the body's circardian rhythm may affect normal physiological function including blood pressure, cardiac output, gastric acid secretion, enzymatic activity, blood flow, body temperature. Thus these vital functions are regulated by transient release of active drug or substance at specific time and site. Pulsatile drug delivery systems are designed according to circardian rhythm of the body. These systems deliver the drug at specific time and specific site as per the pathophysiological need of the disease. In view of emerging chronotherapeutic approaches, pulsatile drug delivery systems are gaining a lot of interest now days. Floating Pulsatile drug delivery system (FPDDS) concept was applied to increase the gastric residence time of the dosage form having lag phase followed by burst release. To overcome limitation of various approaches for imparting, buoyancy and lag controlling were prepared by floating Pulsatile delivery system, for which time controlling system like rupturable membrane, erodible coating, capsule shaped system or multiparticulate system are primarily used in the control release.
