AbstractPlatelets play an important role in haemeostasis and other thrombotic disorders but they may also be involved in other physiological and pathological processes. Many similarities have been found between the central nervous system and platelets. Moreover, due to the absence of nuclei, platelets provide a suitable simplified model for the study of signal-dependent protein transcription and programmed cell death. The goal of this study was to identify the most efficient and replicable method of sample preparation for proteomic analysis of platelets isolated directly from peripheral blood. This method was intended for application to biomarker screening. Emphasis was put on different centrifugation forces and lysis solutions on protein concentration, method complexicity, repeatability, and compatibility with mass spectrometry.
