The invention discloses a method for determining ester compounds in tobacco through supercritical fluid chromatography (SFC)-gas chromatography-mass spectrum (GC-MS). According to the method, supercritical fluid is used for extracting ester compounds in tobacco, the extraction fluid is divided into a plurality of segments of fractions through the SFC, and each fraction is analyzed by the GC-MS. The compounds of the separated fractions cannot be overlaid basically, and the problem that the peak capacity of the GC-MS is limited is solved. The result is satisfactory, the number of the separated ester compounds is much larger than that of direct injection analysis, and a novel method is provided for determining ester compounds in tobacco.
