重组Clostridium thermosulfurogenes β-淀粉酶的分泌表达

为将Clostridium thermosulfurogenesβ-淀粉酶基因高效分泌表达,构建了带有omp A信号肽的表达载体和重组大肠杆菌。通过测定酶活力考察诱导物浓度、诱导时相、表面活性剂对重组菌分泌表达β-淀粉酶的影响。结果omp A信号肽介导Clostridium thermosulfurogenesβ-淀粉酶分泌率为23.6%,略优于pel B信号肽;重组菌生长的对数前期和100μmol//L的IPTG浓度是较优的诱导表达条件;重组菌诱导表达初期添加1%吐温80,β-淀粉酶分泌酶活力提高了105%,而在诱导至较高细胞密度后添加0.05%Triton X-100,β-淀粉酶分泌酶活力可提高301%。
