Screening the Peptides Specifically Binding the Injured Endothelial Cell with Two Phage-Displayed Random Peptide Libraries

Objective:To screen the peptides specifically binding the injured endothelial cell(EC) by means of both 6-mer and 15-mer peptides libraries displayed on the surface of filamentous bacteriophage(FUSE5).Methods:Cultured calf aortic endothelial cells were injured by incubating with oxidized low density lipoprotein(ox-LDL) for 4 hours.The binding phage to injured EC was obtained by three rounds of screening and the inserted amino acid sequences of displayed peptides was confirmed by DNA sequences.Results:We primarily isolated ten phage clones from two phage display libraries.Analysis of peptide shows that eight clones include 6-mer sequence and two clones contain 15-mer sequence.Conclusion:We obtained the peptides specifically binding with high avidity to injured EC by the phage-displayed approach.
