The preparation method comprises the following steps: (1) preparing pellets from glucose and drying; (2) putting the dried pellets into a fluidized bed for fluidizing; and (3) enveloping the surfaces of the pellets in the fluidized bed with melt fatty alcohol and/or fatty acid, to prepare the bypass rumen protective glucose granules. The bypass rumen protective glucose can more efficiently pass through the bypass rumen of ruminant animals, the availability of the enveloped layer by the ruminant animals is increased, and the enveloped glucose can be completely released and fully utilized by the ruminant animals, so that the ketosis of the ruminant animals in perinatal period can be effectively prevented, the occurrence of ketosis or subclinical ketosis can be decreased, the postnatal weight loss can be reduced, the nonreturn rate can be improved, and the milk yield can be increased.
