Effects of Fujian Tablets on the Expression of GFAP and rCBF in the Brains of MCAO Model Rats

Objective:To investigate the correlation of the changes of the expression of GFAP with rCBF in the brains of MCAO rat models affected by Fujian tablets.Methods:72 healthy male Wistar rats were randomly divided into three groups including sham operation group,model group and administration group.Middle cerebral artery occlusion models were established by electrocoagulation.The solution was Fujian tablets was administrated in administration group and equal volume of normal saline was administrated in sham operation group(P0.01)and model group by gavage for 2 weeks respectively.Laser Doppler Flowmetry(LDF)was used to measure rCBF in lesioned frontal lobe cortex.Immunohistochemistry was used to observe the expression of GFAP in the lesioned cortex.Linear correlation analysis was used to analyze the correlation of changes of the expression of GFAP with those of rCBF.Results:After operation,rCBF in the lesioned frontal lobe cortex of the rats of model group and administration group both decreased remarkably compared with sham operation group(P0.05).But after administration of Fujian tablets for 1 week,rCBF in the lesioned frontal lobe cortex increased remarkably(P0.05,vs before administration)and is also more than in model group in synchronization(P0.05),though it is still lower than that in sham operation group.After administration for 2 week,rCBF is still more than that in model group in synchronization.In model group little changes occurred in rCBF as well as in sham operation group.Immunohistochemistry showed after operation little difference in the expression of GFAP exists between these three groups(P0.05).After administration of Fujian tablets for 1 week and 2 weeks,both the expressions of GFAP enhanced compared with other two groups in synchronization and administration group 1 week after operation(P0.05).Linear correlation analysis demonstrated rCBF is closely related to the expression of GFAP in the administration group after administration for 1 week and 2 weeks.Conclusion:Fujian tablets can improve the activity of astrocytes in brains of model rats,which can facilitate increasing rCBF in lesioned cortex.
