The driving need for productivity improvement in drug discovery and development has accelerated the development of advanced cellular models that better mimic the in vivo physiology of living tissues and organisms. The past decades have witnessed an increase in the number of 3D cell culture technologies for drug discovery, including 3D culture gels, scaffolds, bioprinting, organoids, and organs-on-chips. This chapter provides an overview of leading 3D cell culture and dish based organogenesis technologies, and discusses their implications, and limitations, for drug discovery.
