Determination of gossypol in cotton seed kernel in southern Xinjiang by high performance capillary electrophoresis

Objective High performance capillary electrophoresis(HPCE) method was established for the separation of gossypol in cotton seed kernel.Methods The separation was carried out on a fused-silica uncoated capillary column (58.5 cm ×75 μm).The sample was injected by hydro-dynamic injection(0.5 Pis×10 sec) at a temperature of 25℃.The analysis was completed in 0.01M sodium tetraborate-phosphoric acid buffer(pH8.00) at 20 kV of separation voltage and 235 nm of detection wave length.Results The linear range of gossypol was 5.3~49.8 μg/ml and the correlation coefficient was 0.999 8.The limits of quantitation were 0.08 μg/ml The average recoveries were in the range of 97.5%~98.4%.Gossypol content of different cottonseed in Southern Xinjiang was 0.60%~1.04%.Conclusion The method has good precision and recovery.It can be used for determination of gossypol in cotton seed kernel.
