Expression and significance of Nogo-A in PC12 cells differentiation

Objective To explore the expression and significance of NOGO-A in PC12 cells differentiation.Methods PC12 cell,a rat's phaeochromocytoma cell line,was cultured and was induced by nerve growth factor(NGF).The proliferation and neurite extension of PC12 cells was observed by inverse microscopy.The expression of NOGO-A in PC12 cell differentiation was detected by immunofluorescence staining,semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR)and Western blot method.Dopamine(DA)level in the culture medium was detected on the 1 st,3 rd,5 th and 7 th days.Results NOGO-A was not expressed in undifferentiated PC12 cells.In PC12 cells induced by NGF,with cell axon extension,the expression of NOGO-A increased gradually(P0.05).NOGO-A,which was not expressed in undifferentiated PC12 cells,increased during PC12 differentiating into sympathetic neuron(P0.05),but dopamine(DA)level presented little difference.Conclusion During PC12 differentiating into sympathetic neuron,the expression of NOGO-A increases gradually,presumed that NOGO-A may enhance neurite growth during early development of neuron,but has little regulation effect on dopamine secretion.
