Study on inhibition of proliferation and induction of apoptosis in BGC-823 cell by cordycep militaris.

Objective To study the effects of the aqueous extract from Cordycep Militaris(AEoCM) on proliferation and apoptosis of BGC-823 cells.Method BGC-823 were incubated with AEoCM at different concentrations for 24 h,48 h and 72 h respectively.The morphology of BGC-823 cell was observed under a light microscope;The effects of AEoCM on cell proliferation were measured by MTT colormetric assay;Flow Cytometry(FCM) was used to detect cell cycle distribution and apoptotic rates;The expression of NK-kBP65 was examined by the immunohistochemistry method.Result AEoCM inhibited the proliferation of BGC-823 cells in a time and dose-dependent manner.After exposure to cordycepin,the typical morphological features of apoptosis were observed,including cell shrinkage and darkness.The cell cycle of BGC-823 was apparently arrested in G2 phases by AEoCM in time-dependent manner.The apoptotic rates of 0.5 g/L AEoCM in 24 h,48 h and 72 h time intervals were 4.655%,11.039% and 19.368% respectively,and the expression of NK-kBP65 decreased significantly.Conclusion AEoCM inhibits the proliferation of BGC-823 cells and induces apoptosis,which may be associated with the down regulation of the expression of NK-kBP65.
